What it is
Humanin is a 24-amino-acid peptide encoded within the mitochondrial 16S rRNA gene. It was discovered for its ability to protect neurons from amyloid-beta toxicity. It signals through multiple receptors and pathways, including gp130/IL-6 receptor family members and the formyl peptide receptor-like 1.
What the evidence shows
Preclinical studies show neuroprotective, anti-apoptotic, and metabolic effects. Humanin levels decline with age and certain diseases, and higher circulating humanin is associated with longevity in some cohorts. Human interventional trials are lacking.
Risks & unknowns
Human safety data are absent. Because humanin affects survival signaling and metabolism, off-target effects are plausible. Unregulated product quality is a concern.
