What it is
Gonadorelin is a 10-amino-acid decapeptide produced in the hypothalamus. Pulsatile release stimulates LH and FSH secretion, which drive gonadal steroid production. Synthetic gonadorelin is used as a diagnostic agent and in some assisted-reproduction protocols.
What the evidence shows
Gonadorelin has extensive human evidence for diagnosing central hypogonadism and for stimulating ovulation in fertility treatment. Continuous administration paradoxically suppresses the reproductive axis (the basis for GnRH agonist drugs used in prostate cancer, endometriosis, and IVF downregulation).
Risks & unknowns
Pulsatile use can cause ovarian hyperstimulation, multiple pregnancies, and local reactions. Continuous use causes an initial flare followed by suppression of sex hormones, with hot flashes, bone loss, and mood changes. Use requires specialist oversight.
Legal / regulatory status
Gonadorelin is FDA-approved for diagnostic evaluation of gonadotropic function. Related GnRH agonists and antagonists are widely approved for oncology and fertility. Gonadorelin is prohibited by WADA under S2.
