What it is

GHK-Cu is a copper-binding tripeptide: the amino-acid sequence glycyl-L-histidyl-L-lysine (GHK) complexed with a copper(II) ion. It occurs naturally in human plasma, saliva, and urine, and its concentration declines with age. Because copper is essential for building collagen and elastin, GHK-Cu is often framed as delivering copper to connective tissue while itself signaling repair.

At the cellular level, reviews describe GHK-Cu stimulating fibroblast activity and collagen synthesis, promoting blood-vessel and nerve outgrowth, and increasing growth factors such as VEGF and bFGF at low (nanomolar) concentrations. A 2018 review reports that a striking fraction of human genes — about 31.2% — showed expression changes of 50% or more in the presence of GHK. Read that carefully: it is an in-vitro gene-expression observation, evidence of mechanism, not a demonstrated clinical result.

What the evidence shows

GHK-Cu is the one peptide on this list with a legitimately human evidence badge — with an important caveat: the human data are overwhelmingly topical and cosmetic. Multiple small controlled facial-skin trials (roughly 40–70 women each, typically over 12 weeks) reported that copper-peptide creams improved skin laxity, clarity, density, and thickness and reduced fine lines and wrinkles, in several cases outperforming comparator creams such as vitamin C or retinoic acid. One biopsy analysis found collagen increases in 70% of women treated with GHK-Cu versus 50% with vitamin C and 40% with retinoic acid.

Everything beyond skin cosmetics — wound healing, tissue protection, anti-fibrotic and "regenerative" effects — rests mainly on cell-culture and animal studies (rabbits, rats, mice). So the honest framing is: solid, if modest and small-scale, human evidence for skin appearance; mechanistic and preclinical evidence for the bigger regenerative claims. The human tier reflects the cosmetic trials; it does not certify the systemic anti-aging story.

Risks & unknowns

Topically, GHK-Cu has a long cosmetic track record and the reviewed literature reports it as well tolerated, with no significant safety issues documented in its cosmetic and wound-healing use. What is not established is the safety or efficacy of injected or high-dose systemic GHK-Cu, which the human cosmetic trials did not test. Product quality also varies: peptide stability, purity, and actual skin penetration are recurring limitations noted in the literature, and results depend heavily on formulation.

Legal / regulatory status

GHK-Cu (as copper tripeptide-1) is regulated as a cosmetic ingredient and is widely and legally sold in skincare. It is not an FDA-approved drug for any medical indication, and no prescription GHK-Cu product has received approval. Cosmetic status governs topical use for appearance; it does not authorize marketing GHK-Cu as a treatment for any disease.

That cosmetic status covers topical products only. In September 2023 the FDA placed injectable GHK-Cu specifically in Category 2 of its bulk drug substances list — the same significant-safety-risk category as BPC-157 and the thymosin-β4 fragment (TB-500) — effectively barring compounding pharmacies from making injectable GHK-Cu for human use. That lines up with the caution above: injected or systemic GHK-Cu is neither approved nor endorsed here.
